学术报告―Transcriptional and post-transcriptional regulation of gene expression

Gene expression is the most fundamental mechanism to dictate cell function. The regulation of gene expression has known to be processed on multiple levels, including chromatin signatures, transcription, post-transcriptional modification, translation and mRNA degradation. In this presentation, I will discuss two steps of gene regulation: transcriptional initiation and post-transcriptional regulation. In the first part, by using recent published CHIP-seq data in CD4+ T helper (Th) cells, we computationally identified "super enhancers" as combination of contiguous binding sites of master transcription factors. The super enhancers are cell type specific and associated with signature genes, which demonstrate their importance in Th cell differentiation. In the second part, we investigated the miRNA regulation on mRNA degradation and protein production. We found that in addition to down-regulating mRNA levels, miRNA also directly repress translation of hundreds of genes. Furthermore, we showed that one miRNA (let-7) can tune protein synthesis from thousands of genes.
