法国巴黎2019生命科学暑期研学营日志-2019.07.10

   法国时间7月10日,今天是在夏令营学习的第三天,我们来到了ICSN(Natural Pro-duct Chemistry Institute)。首先来自法国国家科学研究院(CNRS)的研究员Litaudon老师为我们做了一场关于如何从天然植物中发现、筛选新药物的报告。

   Litaudon老师首先为我们介绍了ISCN的规模及其主要的研究方向。并且很系统地介绍了药物筛选和发现的科研背景、研究现状以及当前面临的挑战。老师为我们列出了原有研究方法的局限性和问题,而且与我们分享了分子网络(Molecular Networking)这项新方法。其中老师所讲的一句话令我们印象深刻——“当你不知道如何着手开始一项发现药物的项目时,大自然会是一个好的出发点”。

  随后,和蔼可亲的Touboul老师主要为我们讲解了如何利用质谱法对天然或合成产物进行结构分析,并带领我们参观了他的实验室,介绍了质谱仪、气相色谱仪等仪器,以及相关的实验方法等。两位老师讲授知识时都深入浅出,努力用精确的描述消除我们之间由于语言不通造成的些许交流障碍。并将原理、应用技术和研究前景相结合,为我们队伍带来了一场汲取新知识的视听盛宴。

  中午,我们体验了法国当地正宗的菜肴,地道制作,纯正美味。法国传统菜肴有十三道菜可供选择,因此每道菜分量不大,但是相当精美。如今一切从简,我们此次品尝的就是包括三道菜的法餐,前后顺序为:开胃菜,主菜,甜品。开胃菜中,摆放着蘸有果醋的酸爽脆嫩的生菜,加上一块中间放有酱汁的羊奶酪,配以一块香脆的切片面包。接下来就是大家十分期待的主菜,或许是法国人对于中国朋友的优待,除了鲜嫩的巴沙鱼与蔬菜,还有带有奶香味的米饭,半颗撒上香料的熟番茄,一片调味的柠檬。最后是甜品,是一小杯草莓沙拉,上面点缀着一小株薄荷叶,给酸甜的草莓增添了一抹清凉。除此之外,餐桌上还有可以自取的面包和气泡水。透过餐厅的落地窗,窗外树枝摇曳,阳光洒落在餐桌上,大家在白色长桌上尽情享受美食,有说有笑。

  下午,我们首先听了PINET老师关于NMR(核磁共振)的简单报告,让我们了解到了通过核磁共振,如何获取生物大分子的原子级别的信息。在她的带领下,我们还近距离参观了核磁共振测定的仪器。

  之后,我们去往凡尔赛大学,参观了他们的遗传学和细胞生物学的实验室。我们先是了解了学院的概况和研究主题,接着就是对部分实验室进行参观学习。

  首先去到的是遗传学实验室,这里可以说是我们最有亲切感的地方了。在这里我们见到了不同类型但是十分熟悉的PCR仪,以及自己平时实验经常使用的移液枪。

  接下来我们参观了质谱仪和流式细胞仪。这些仪器与我们通常了解到的有些许不同。流式细胞仪与荧光3D成像相结合,能够让细胞的筛选与观察同时进行。

  最后,VINIT老师为我们介绍了果蝇在帮助理解人类疾病过程中发挥的作用。果蝇主要有以下优点,后代多,价格低廉,基因型丰富,并且有大量与一些哺乳动物相同或相似的基因和信号通路,目前已有6项诺贝尔奖是与果蝇相关的。

  在结束晚餐回到住处的林荫路上,我们遇见了一群打乒乓球的法国朋友,不禁走上前去与他们切磋。尽管语言不通,但我们可以以球会友,场面一度十分欢快。






  Today is the third day of our summer school. We came to ICSN(Natural Product Chemistry Institute).At first, Mr. Litaudon did a presentation on how to select new drugs from nature, regarding biodiversity as a source of new drug discovery.

  Mr. Litaudon first introduced the scale of ISCN and its main research directions to us. He also introduced the research background, research situation and current challenges of drug discovery systematically. The teacher listed the limitations and problems of the original research methods for us, and shared us the new method ‘Molecular Networking. Moreover,he emphasized an attitude towards drug discovery ,when you have no idea where to begin in a drug discovery program, nature is a good starting point.After this short but abundant presentation, he also showed us his laboratory that stores natural plants.

       

  After that, Mr. Touboul did a presentation on structural characterization of natural and synthetic products by Mass Spectrometry. He led us to visit his laboratory and introduced instruments such as mass spectrometers and gas chromatographs, as well as relevant experimental methods. Both of these two teachers made the knowledge easy to understand, trying to eliminate the communication barriers between us by a precise description. They combined principles, application techniques and research prospects together.

   

  At noon, we tried authentic local dishes in France.There are 13 different diches in French traditoinal diches that you can choose, so each dish is not very large, but it is quite awesome. Now everything is simple, we are having a three-course French meal, the order is: appetizer, plat, dessert.For the appetizer, there was tender lettuce with vinegar, and a piece of g-oat cheese with sauce in the middle, served with a crisp sliced bread. And then,the plat was which everyone was looking forward to, thanks to the French people's sympa treatment for the Chinese friends, in addition to the fresh basha fish and vegetables, there was rice with milky aroma, half a spiced tomato,and a piece of seasoned lemon . Finally, the dessert was a small cup of strawberry salad with a small mint leaf on it. There were also some bread an-d sparkling water on the table. Through the floor-to-ceiling windows of the restaurant, the branches swayed outside, and the sun shone on the table. Everyone enjoyed the food on the long white table, talking and laughing.

   

  In the afternoon, we first listened to Miss PINET about NMR (nuclear magnetic resonance) , made us understand how to obtain the information of biological macromolecules in the atomic level through nuclear magnetic resonance .Under the guidence of her,she show us to the NMR spectrometer closely.

   

  Then we went to the university of Versailles-Saint-Quentin and visited their laboratories of genetics and cell biology.We first learned about the presentation of the faculty and overview of research themes and visited some laboratories.

  The first place to go is the genetics lab, which is arguably the most intimate place for us.Here we saw different but very familiar PCR machines and pipettes that we often used in our daily experiments.

   

  Next we visited the mass spectrometer and the flow cytometer.These instruments are slightly different from what we usually know.The flow cytometer combined with fluorescence 3D imaging make it possible to select and observe cell simultaneously.

   

  Finally, Mr VINIT gave us an introduction to the role that Drosophila plays in helping people to understand human disease.Drosophila have some advantages for genetics,for example: large progeny, inexpensive, numerous genotypes, and numerous genes and signalin pathways in some mammals. Currently, there are 6 Nobel prizes related to Drosophila.

   

  After dinner, on the tree-lined road back to the residence, we met some French friends who played table tennis.Although we can not speak French, we can play table tennis with each other, and we were very happy.
