At chemical synapses, the spatial correlation between the active zone, where vesicles fuse, and the distribution of post-synaptic receptors, which sense the neurotransmitter, is one of the critical parameters that shape synaptic strength. Using electron microscopy tomography, we have able to identify novel structures involving the proteins UNC-10/RIM and SYD-2/liprin that localize synaptic vesicles at the active zone of cholinergic neuromuscular junctions. On the post-synaptic membrane, ionotropic acetylcholine receptors (AChRs) are clustered at high density in front of ACh release sites. Using a forward genetic strategy, we identified 3 proteins required for AChR clustering that form on an extracellular scaffold within the synaptic cleft.
